Effect and Meta-Analysis of Huoxuedingxuan Capsule Containing Serum on Proliferation, Migration and VEGF Expression of Rat Endothelial Progenitor Cells

Abstract
To study the effect of serum containing Huoxue Dingxuan Capsule on migration and tube formation of EPC cells in rats, and to explore the mechanism of Huoxue Dingxuan Capsule in treating CSA and its effect on angiogenesis. Methods: Rat EPCs cultured in vitro were identified by double fluorescence staining and divided into four groups: blank control group, low dose group, middle dose group and high dose group. The blank control group used M199 complete medium containing 10% fetal bovine serum, VEGF10ng/ml and bFGF4ng/ml, and the low, middle and high experimental groups were added with 10% different concentrations of drug-containing serum. After 3 days of culture, flow Edu cells were detected. Results: The results of flow Edu showed that the number of positive cells in blank control group, low dose group, medium dose group and high dose group were 11.43%, 49.99%, 30.01% and 21.21% respectively. Compared with the control group, the serum containing different concentrations of Huoxue Dingxuan Capsule can up-regulate the expression level of VEGF protein and mRNA, and the expression level of VEGF protein and mRNA in the low dose group is significantly higher than that in the high and middle dose groups (P<0.05). Conclusion: The serum containing Huoxue Dingxuan Capsule can promote the proliferation of EPCs, and the low-dose group with 10% concentration of serum has the best effect than other groups, which may promote the angiogenesis by regulating the expression level of VEGF. And through meta-analysis, we found that serum containing Huoxue Dingxuan Capsule can interfere with mouse brain microvascular endothelial cells and reduce autophagy.
