Guben Zenggu Granules Restore MC3T3-E1 Cells from H2O2- Induced Oxidative Damage

Abstract
Oxidative stress is an imbalance in the production and elimination of reactive oxygen species in the body. It is accompanied by cell and tissue damage, and the accumulation of oxidation products can cause disease. This study focused on the effects of H2O2 on the antioxidant activity, cell proliferation and protein gene expression levels in MC3T3-E cells, and explored the repair effects of Guben Zenggu Granules on H2O2 -induced oxidative damage. By culturing MC3T3-E1 cells in experimental medium, establishing H2O2 cell induction model and culturing serum containing various concentrations of Guben Zenggu Granules, compare the cell ALP content, MDA, SOD and LDH activities, as well as Coll, NOX4, OC, OPG, RANKL, Runx2/Cbfa1 related gene proliferation rate and relative gene expression level. The decomposition of H2O2 is also measured using Ca+ concentration. Guben Zenggu Granules significantly decreased ROS value, cell MDA content, SOD activity and LDH concentration, Runx2, OPN, ColⅠ, OPG, OC, Nox2 and other related gene protein expression and indicators tended to normal values. The results show that Guben Zenggu Granules can play an important role in restoring the functions of MC3T3-E1 cells from H2O2 -induced oxidative damage.
